Before looking at the generator, list what is different from the qualified state:

Load — new equipment, more consumables, cardboard packaging, a cabinet left closed.

Space — a new penetration, a damaged door seal, a modified HVAC damper, a new cold surface.

Season — higher ambient humidity in summer, colder walls in winter.

Equipment — sensor calibration, desiccant condition, catalyst age, injection system maintenance.

Indicators — a new BI lot or supplier, a change in incubation, transport or handling.

Cycle records make this comparison quick: overlay the current trend of humidity, concentration and temperature on a qualified cycle and look for where they diverge.

Symptom → likely causes

Symptom

Likely causes

What to check

Dehumidification target not reached, or takes much longer

Air leaking in from outside the boundary; HVAC not fully isolated; high ambient humidity; wet surfaces or liquids in the space; saturated desiccant

Seals and dampers; ambient RH compared with qualification; desiccant regeneration; remove liquids

Concentration does not reach set point, or falls during exposure

Leaks; absorbent materials taking up vapour; surfaces that decompose peroxide (some metals, organic residues); injection rate lower than expected; sensor drift

Leak test (isolators) or smoke/detector check at seals (rooms); load list; injection system and solution; sensor calibration

Condensation alarm or visible droplets

Cold surfaces; overdosing for the temperature and humidity; temperature falling during the cycle; vapour jet aimed at a nearby surface

Surface temperatures; dose and set points; generator and fan positions; HVAC temperature control

BI growth at one or a few locations

Poor distribution to that point; shadowing by equipment; folds (gloves, bags); absorbent material nearby; cold spot

Distribution — fan positions, airflow; what was near the BI; chemical indicator result at the same point

BI growth scattered across unrelated locations

BI lot quality; damaged or wet BIs; handling or transfer contamination; incubation conditions; occasional “rogue” BI

BI certificate and lot; handling and transfer; positive controls; repeat with a fresh lot

Aeration much longer than qualified

Absorbent materials releasing peroxide; reduced exhaust or recirculation flow; ageing catalyst; sensor reading high

Load list; airflow and fan speeds; catalyst condition; sensor calibration

H₂O₂ detected outside the treated space

Door seals; dampers not closed; cable and pipe penetrations; pressure regime of the room

Low-range detector survey around the boundary; damper positions; room pressure during the cycle

Results vary from day to day

Ambient humidity and temperature; load not standardised; door opening during preparation; operator steps not fixed

Ambient records; written load configuration; procedure adherence

Reading biological indicator failures

The pattern of positive indicators is the most useful clue. Growth concentrated in one area — always behind the same equipment, or always at glove fingertips — says the vapour is not reaching that place at the required dose. Changing the cycle globally may hide the problem without fixing it; changing the distribution or the configuration at that point fixes it.

Growth scattered without a pattern, with chemical indicators at the same positions showing full exposure, points to the indicators rather than the process. Occasional isolated positives — “rogue” BIs — are a known limitation of biological indicators in vapour processes.
