For oxidation-sensitive drugs, especially biologics, the question is not only "is the air below 1 ppm?" but "how much peroxide ends up in the container?" This guide explains where uptake comes from and how to control it.

Fenggu VHP engineering team4 min readUpdated October 2026

Key points

After aeration, low levels of H₂O₂ remain in the isolator air and continue to off-gas from gloves, gaskets and plastics.

Open containers and liquid product exposed to that air can pick up peroxide during filling; the amount depends on concentration, exposure time and surface area.

Oxidation-sensitive products — many proteins, peptides and some small molecules — may be affected at very low levels.

The aeration endpoint for filling should come from a product uptake study, not only from the 1 ppm occupational limit.

Design controls (low-absorbing materials, closed product path, good aeration capacity) usually do more than simply aerating longer.

Two different limits

When an isolator is decontaminated with vaporized hydrogen peroxide, the cycle ends with aeration: the vapour is removed until the concentration falls below a set point. That set point is often borrowed from occupational exposure limits — 1 ppm is the common figure — because it is what makes the isolator safe to open and to work at.

For filling, the relevant question is different. The product is not breathing the air; it is sitting in open vials, syringes or bags, exposed to that air for as long as the filling run lasts. Hydrogen peroxide is highly water-soluble, so even a very low concentration in the air can transfer into an aqueous product. For a robust small molecule this rarely matters. For an oxidation-sensitive biologic, it can.

Where the residual peroxide comes from

  <rect x="10" y="10" width="740" height="300" rx="12" fill="#F5F8FB" stroke="#DCE3EC"/>
  <rect x="40" y="40" width="420" height="250" rx="8" fill="#FFFFFF" stroke="#5A6B82" stroke-width="2"/>
  <rect x="60" y="48" width="380" height="18" rx="3" fill="#C9D3DF"/>
  <text x="250" y="61" font-size="11" fill="#2B3A4F" text-anchor="middle">HEPA supply</text>
  <g stroke="#0A8F8A" stroke-width="1.2" stroke-dasharray="3 4"><line x1="110" y1="72" x2="110" y2="240"/><line x1="200" y1="72" x2="200" y2="240"/><line x1="300" y1="72" x2="300" y2="240"/><line x1="390" y1="72" x2="390" y2="240"/></g>
  <circle cx="40" cy="150" r="20" fill="#E6EDF4" stroke="#5A6B82" stroke-width="2"/>
  <circle cx="40" cy="220" r="20" fill="#E6EDF4" stroke="#5A6B82" stroke-width="2"/>
  <rect x="80" y="240" width="340" height="12" fill="#C9D3DF"/>
  <rect x="100" y="200" width="70" height="40" rx="4" fill="#E9DFC9" stroke="#B49A63"/>
  <text x="135" y="224" font-size="10" fill="#5B4A1F" text-anchor="middle">stoppers</text>
  <g fill="#FFFFFF" stroke="#2B3A4F"><rect x="230" y="210" width="16" height="30"/><rect x="252" y="210" width="16" height="30"/><rect x="274" y="210" width="16" height="30"/><rect x="296" y="210" width="16" height="30"/></g>
  <rect x="340" y="150" width="8" height="55" fill="#5A6B82"/>
  <rect x="330" y="140" width="28" height="12" fill="#5A6B82"/>
  <rect x="380" y="195" width="60" height="45" rx="3" fill="#FFFFFF" stroke="#2B3A4F" stroke-dasharray="4 3"/>
  <text x="410" y="222" font-size="10" fill="#2B3A4F" text-anchor="middle">tub lid</text>
  <g font-size="12" font-weight="700" fill="#FFFFFF">
    <circle cx="40" cy="120" r="11" fill="#D62B2B"/><text x="40" y="124" text-anchor="middle">1</text>
    <circle cx="80" cy="280" r="11" fill="#D62B2B"/><text x="80" y="284" text-anchor="middle">2</text>
    <circle cx="135" cy="190" r="11" fill="#D62B2B"/><text x="135" y="194" text-anchor="middle">3</text>
    <circle cx="410" cy="185" r="11" fill="#D62B2B"/><text x="410" y="189" text-anchor="middle">4</text>
    <circle cx="372" cy="135" r="11" fill="#D62B2B"/><text x="372" y="139" text-anchor="middle">5</text>
  </g>
  <g font-size="13" fill="#0E1A2B">
    <text x="490" y="60" font-weight="700">Typical sources</text>
    <text x="490" y="95">1  Gloves and sleeves</text>
    <text x="490" y="125">2  Gaskets, seals, plastic panels</text>
    <text x="490" y="155">3  Stoppers and components in bowls</text>
    <text x="490" y="185">4  Nested tubs with permeable lids</text>
    <text x="490" y="215">5  Product path and filling needles</text>
    <text x="490" y="255" fill="#5A6B82" font-size="12">All absorb vapour during gassing</text>
    <text x="490" y="273" fill="#5A6B82" font-size="12">and release it during aeration</text>
    <text x="490" y="291" fill="#5A6B82" font-size="12">and filling.</text>
  </g>

Schematic of a filling isolator. Numbered points are the usual places peroxide is absorbed and later released.

Aeration removes peroxide from the air quickly, but materials that absorbed it during the gassing phase keep releasing it for much longer. The concentration in the air at the start of filling, and its slow decline during filling, are governed largely by these materials:

Elastomers — gloves, sleeves, door and window gaskets, inflatable seals.

Plastics — panels, guards, change parts, bags and tubing.

Components loaded before the cycle — stoppers, caps and especially nested tubs. Lids made of spunbonded polyolefin are permeable to the vapour, so the inside of the tub, and the containers in it, may be exposed.

Product-contact parts — if the filling needles, tubing or pumps are open to the isolator during the cycle, peroxide can be adsorbed on surfaces that later contact the product directly.

Which products are at risk

Hydrogen peroxide is an oxidant. Proteins are the classic sensitive case: residues such as methionine and tryptophan can be oxidised, which may affect potency, aggregation or immunogenicity. Published studies on monoclonal antibodies and other biologics have shown measurable oxidation after exposure to peroxide at very low concentrations in solution, and some small-molecule APIs and excipients (for example polysorbates) are also sensitive. Whether a given product is affected has to be established for that product; there is no universal safe level.

How to measure uptake

An uptake study answers the question directly: how much peroxide does the product, or a surrogate, pick up under worst-case conditions?

Element

Typical approach

Surrogate

Water for injection or placebo buffer in the real container, filled at the start of the run when residual levels are highest

Positions

Open containers at worst-case locations: near gloves, near absorbent materials, at the start of the conveyor, low-airflow spots

Exposure

The longest realistic open time, including line stoppages

Assay

A method sensitive enough for the expected range — usually a fluorometric or other low-level peroxide assay, validated for the matrix

Air data

Low-range H₂O₂ sensor readings in the isolator during the study, so air concentration can be related to product uptake

Product impact

For sensitive products, a spiking study showing which concentration in solution starts to affect quality attributes

Combining the two results gives a defensible link: the air concentration at which filling may start, and the margin between expected uptake and the level that affects the product.

Controls that work

Lever

What to do

Aeration endpoint

Set the filling start condition from the uptake study. For sensitive products this is often well below 1 ppm, which needs a low-range sensor to verify.

Aeration capacity

Sufficient air changes and a catalytic converter in the recirculation or exhaust path shorten aeration; warmer air helps materials release peroxide faster.

Materials

Prefer low-absorbing materials inside the isolator (316L, glass, PTFE, PVDF); minimise exposed elastomer and soft plastic area. See VHP material compatibility.

Closed product path

Keep pre-sterilised product-contact parts closed or protected during the cycle so they are not exposed to vapour.

Component loading

Decide what is in the isolator during gassing. Components introduced afterwards through a decontaminated transfer route are not loaded with peroxide.

Cycle dose

Avoid overdosing. A cycle developed to deliver the required lethality with margin, rather than the maximum, leaves less to aerate.

Fill sequence

Where justified by data, discard or segregate the first containers filled, when residuals are highest.

Most of these are design decisions. They are far easier to make in the URS than after the isolator is built, which is why product sensitivity to peroxide should be one of the first questions asked when a filling isolator is specified.

For the overall approach to isolator selection see how to choose a pharmaceutical isolator; for cycle development and qualification see VHP cycle development and IQ/OQ/PQ on our main site.

References

EU GMP Annex 1 (2022), Manufacture of Sterile Medicinal Products

PDA Technical Report No. 34, Design and Validation of Isolator Systems for the Manufacturing and Testing of Health Care Products

ISO 13408-6, Aseptic processing of health care products — Isolator systems

ISO 22441:2022 — Low temperature vaporized hydrogen peroxide sterilization

  Equipment
Fenggu equipment for this

FG-ISO-AF
Aseptic filling isolator

Integrated VHP bio-decontaminates exposed interior surfaces before every campaign; unidirectional airflow, H14 HEPA, pressure and particle monitoring.
View specifications →FG-ISO-CC
Cell processing isolator

For open handling of cells and biologics, where peroxide sensitivity is often a key requirement.
View specifications →FG-PB-VHP
VHP pass box

Transfers components into the isolator after the main cycle, so they are not loaded with peroxide during gassing.
View specifications →

Related application guides: Isolator and RABS decontamination · Cell and gene therapy

  FAQ
Common questions

<details>
Is 1 ppm H₂O₂ low enough to start aseptic filling?

1 ppm is an occupational exposure limit, not a product limit.
