Many sites first produce a list of existing procedures. This guide explains what Annex 1 asks for, a structure that works, and how to keep the CCS alive after it is written.

Fenggu VHP engineering team3 min readUpdated October 2026

Key points

A CCS is a site-wide document showing how design, procedures, technology and monitoring together control microbial, particle and pyrogen contamination.

Annex 1 section 2.5 lists 16 elements the CCS should consider, from plant design to continuous improvement.

The useful part is the link from each contamination risk to its controls and to the data that shows the controls work.

It should reference existing documents rather than copy them, and say where gaps remain and what is planned.

A CCS is reviewed periodically and after changes, deviations and trends — it is not written once.

What Annex 1 asks for

The revised EU GMP Annex 1, in force since August 2023, makes the contamination control strategy a central requirement. The CCS is expected to be a planned set of controls, derived from current product and process understanding, covering the whole facility and process. It should define critical control points and assess how effective all the controls — design, procedural, technical and organisational — and monitoring measures are at managing risks to quality and safety.

In practice inspectors look for three things: that the strategy is holistic rather than a collection of separate procedures, that risks are linked to controls and evidence, and that it is used — reviewed, updated and acted on.

The 16 elements in section 2.5

#

Element

What to show

1

Design of the plant and processes, with documentation

How layout, flows and process design prevent contamination

2

Premises and equipment

Cleanroom grades, HVAC, isolators or RABS, equipment design and qualification

3

Personnel

Training, qualification, gowning, behaviour, health and hygiene

4

Utilities

Water, steam, gases and vacuum, their design and monitoring

5

Raw material controls, including in-process controls

Bioburden and endotoxin control of materials

6

Product containers and closures

Preparation, sterilisation or depyrogenation, container closure integrity

7

Vendor approval

Suppliers of key components, sterilisation services and critical materials

8

Management of outsourced activities

Information shared between the parties and responsibilities

9

Process risk assessment

The risk assessments the strategy is built on

10

Process validation

Including aseptic process simulation

11

Validation of sterilisation processes

Terminal sterilisation, filtration, depyrogenation, and decontamination such as VHP

12

Preventive maintenance

Keeping equipment, utilities and premises in their qualified state

13

Cleaning and disinfection

Agents, frequencies, efficacy and residue removal

14

Monitoring systems

Environmental and personnel monitoring, and how the programme is justified

15

Prevention mechanisms

Trend analysis, investigation, root cause, CAPA

16

Continuous improvement

Based on information from the above

A structure that works

Scope and ownership. Products, processes, buildings and grades covered; who owns the CCS and who approves it.

Facility and process description. A short narrative with flow diagrams for people, materials, product and waste.

Contamination hazards. Sources and routes for microorganisms, particles and endotoxins at each process step.

Controls by element. For each of the 16 elements, the controls in place, with references to the procedures, specifications and qualification documents that hold the detail.

Critical control points. The steps where failure would directly affect sterility assurance, and how each is controlled and monitored.

Monitoring and evidence. What data shows the controls are effective, how often it is reviewed and what triggers action.

Gaps and action plan. Known weaknesses, the risk they carry and the planned improvements with dates.

Review. Frequency and triggers for review, and a record of changes.

Linking risk, control and evidence

The most useful part of a CCS is often a matrix: one row per contamination risk, with the controls that prevent it, the monitoring that would detect failure and the document reference. The review should ask whether the controls are still effective, not only whether the document is up to date. Decontamination methods such as VHP are part of the strategy: their qualification, routine records and requalification belong in element 11, and their place in the disinfection programme in element 13 — see cleaning before VHP decontamination.

The regulation itself is summarised in EU GMP Annex 1 for isolators, RABS and cleanrooms.

Barrier technology, decontamination and monitoring all sit inside one contamination control strategy.

References

EU GMP Annex 1 (2022), Manufacture of Sterile Medicinal Products, sections 2.3–2.6

PIC/S PE 009, Guide to Good Manufacturing Practice, Annex 1

ICH Q9(R1), Quality Risk Management

PDA Technical Report No. 90, Contamination Control Strategy Development in Pharmaceutical Manufacturing

 Equipment
Fenggu equipment for this

FG-ISO-F
Aseptic filling isolator

Integrated VHP bio-decontaminates exposed interior surfaces before every campaign; unidirectional airflow, H14 HEPA.
View specifications →FG-PB-VHP
VHP pass box

Integrated VHP with real-time temperature, humidity, pressure and H₂O₂ concentration monitoring and printed cycle records.
View specifications →FG-VHP-C1000 / C2000
HVAC-integrated VHP system

Distributes vapour through the existing AHU to treat a whole suite.
