Inspectors want to see that it works on your surfaces, against your organisms, the way your operators apply it. That evidence comes from a coupon challenge study. This guide covers how to design one: choosing surfaces and organisms, inoculating, applying the disinfectant, neutralising, recovering and calculating log reduction.

Fenggu VHP engineering team3 min readUpdated October 2026

Key points

Coupons of the real surface materials are inoculated, dried, treated as in the SOP, neutralised and recovered.

Use your own environmental isolates as well as reference strains, including spore formers for sporicides.

Log reduction is the difference between recovery from untreated control coupons and from treated coupons.

The neutraliser must be shown to stop the disinfectant without harming the organisms; otherwise results overstate efficacy.

Typical acceptance levels are 3-log for vegetative bacteria and 2-log for spores, defined and justified in the protocol.

Why a site study is needed

Standard tests such as EN 13697 or EN 17126 show that a product works under defined laboratory conditions. They do not cover your surface finishes, your environmental organisms, your dilution, your contact time or the way your operators wipe or spray. USP <1072> describes a site-specific approach based on surface challenge tests, and EU GMP Annex 1 expects the efficacy of disinfection to be demonstrated. The coupon study is that demonstration. The overall programme it supports is described in disinfectant rotation and sporicidal agents.

Study design

Decision

Typical choice

Surfaces

Coupons of each main material in the cleanroom: stainless steel, epoxy or vinyl flooring, glass, polycarbonate, wall panels, curtain material

Organisms

Environmental isolates recovered in your monitoring, plus reference strains — for example a Gram-positive and a Gram-negative bacterium, a yeast, a mould and a spore former

Inoculum

Enough to measure the target reduction with margin — commonly about 105–106 organisms per coupon

Disinfectants

Each product in use, at the in-use dilution and at the end of its stated shelf life

Contact times

The time in the SOP, and often a shorter time to show margin

Replicates

At least three coupons per combination, plus controls

Procedure

Prepare coupons. Cut, clean and sterilise coupons of each material. Size them so the recovery method covers the whole surface.

Inoculate and dry. Spread a known volume of suspension on each coupon and let it dry under controlled conditions. Dried organisms on a surface are harder to kill than organisms in suspension.

Apply the disinfectant. Apply it the way the SOP does — spray, wipe or flood — and leave it for the contact time.

Neutralise. Stop the disinfectant at the end of the contact time by transferring the coupon to a validated neutralising solution.

Recover. Remove survivors by rinsing, swabbing or a contact plate, and enumerate by plate count.

Controls. Treat control coupons in the same way with a non-disinfecting liquid such as sterile water instead of disinfectant.

Calculating log reduction

FormulaLog reduction = log10(Ncontrol) − log10(Ntreated)
N = organisms recovered per coupon. Use the mean of replicates. Two checks are usually made for each disinfectant and organism:

Efficacy: organisms added to disinfectant mixed with neutraliser are recovered at the same level as organisms added to neutraliser alone.

Toxicity: organisms added to neutraliser alone are recovered at the same level as organisms added to a buffer.

A common acceptance is recovery within about 50% (or 0.3 log) of the comparison.

Acceptance criteria

Organism type

Commonly used criterion on surfaces

Vegetative bacteria

≥ 3-log reduction

Bacterial spores (sporicide)

≥ 2-log reduction

Fungi

Defined in the protocol, often 2 to 3-log depending on organism

These values are often taken from USP <1072>.
