Its drawbacks are now hard to justify for routine use. It is classified as carcinogenic to humans, which brings strict controls on exposure, generation and disposal. It needs high humidity and long contact times. Neutralising it with ammonia or ammonium carbonate produces residues, and polymerised formaldehyde can deposit on surfaces, so the room needs cleaning afterwards. Some biosafety laboratories still use it where it is written into established procedures, and in some regions its use as a disinfectant is restricted.

Chlorine dioxide gas

Chlorine dioxide’s strength is distribution. As a true gas it fills complex spaces evenly, reaches into ductwork and does not condense on cold surfaces at normal concentrations. That makes it attractive for large or complicated volumes. It needs high relative humidity to be sporicidal, it is aggressive to some metals and materials, and it is broken down by light, so it is generated on site and its concentration has to be monitored throughout. Exposure limits are low, so sealing and leak monitoring must be rigorous.

Vaporized hydrogen peroxide

VHP has become the default for pharmaceutical and biotech facilities because it combines sporicidal efficacy with a short cycle, no residue and good material compatibility. The process is controlled by the generator — dehumidification, conditioning, the exposure phase and catalytic or ventilated aeration — and the parameters are recorded, which suits GMP validation. Its limitations are real but manageable: absorbent materials such as cardboard and paper take up the vapour and slow aeration, cold surfaces are where condensation starts, and distribution in large rooms needs mixing fans or distribution through the HVAC system.

Which one to choose

Situation

Usual choice

Routine decontamination of cleanrooms, isolators and pass boxes in GMP manufacturing

VHP — short cycle, no residue, validated and recorded

Biosafety laboratory with an established formaldehyde procedure

Often a candidate for replacement by VHP; requires a new validation and a review of materials

Very large or complex volumes, ductwork, poor access for mixing

Chlorine dioxide is sometimes preferred; HVAC-integrated VHP is the alternative where the air handling system can carry the vapour

Rooms with electronics and sensitive equipment left in place

VHP, after confirming material compatibility

Spaces with large amounts of absorbent material that cannot be removed

Remove what you can; whichever agent is used, aeration and efficacy must be proven with the real load

Whatever the agent, the room still has to be sealed, prepared and proven with biological indicators at worst-case locations — see preparing a room for VHP decontamination and biological indicators and the 6-log reduction.

References

EU GMP Annex 1 (2022), Manufacture of Sterile Medicinal Products

EN 17272:2020, Chemical disinfectants and antiseptics — Methods of airborne room disinfection by automated process

US OSHA permissible exposure limits (29 CFR 1910.1000 and 1910.1048)

IARC Monographs Vol. 88 (2006) and Vol. 100F (2012), Formaldehyde

 Equipment
Fenggu equipment for this

FG-VHP-M500
Mobile VHP room sterilizer

Wheeled unit for spaces up to 300 m³, with scheduled and remote start.
View specifications →FG-VHP-C1000 / C2000
HVAC-integrated VHP system

Distributes vapour through the existing AHU to treat a whole suite.
