Real positions are justified by risk assessment and confirmed against the airflow pattern.

Where to sample

Sample locations are chosen by a documented risk assessment and should be representative of the air and surfaces that reach exposed product, without disturbing the airflow that protects it. The smoke study is the best tool for this: it shows where air comes from just before it reaches open containers, and where a probe or plate would sit in its wake.

Particle probes close to the point of fill and the stopper track, pointing into the airflow. Keep the sample tubing to the counter short with few bends, because particles — especially ≥ 5 µm — are lost in long tubing.

Settle plates as close to open product as the process allows, in positions that do not create eddies.

Active air samplers with the head inside and the pump outside, so the isolator’s pressure balance is not upset during sampling.

Surfaces that contact sterile items or are touched during interventions: format parts, guides, the stopper bowl edge.

The VHP problem

Hydrogen peroxide and growth media do not mix well. Three situations need attention.

1. Getting media in without killing them

Plates and sampling heads are usually introduced after the decontamination cycle through a decontaminated transfer route — a VHP pass box, a transfer port, or the isolator’s own airlock. Their outer packaging has to be decontaminated, but the vapour must not reach the agar. Plates designed for this use come in multiple wrappings that are removed one at a time, and the packaging’s resistance to peroxide should be confirmed. Plates that are inside the isolator during the cycle are not valid for monitoring.

2. Residual peroxide on exposed plates

After aeration a low level of hydrogen peroxide remains in the air and continues to off-gas from materials. Settle plates exposed for hours absorb it. At high enough levels it can inhibit the growth of microorganisms that land on the plate — giving a false “no growth”. The defence is a growth promotion test under real conditions: expose plates in the isolator for the full exposure time at the start of a run, then inoculate them with a low number of test organisms and confirm recovery. Some media are supplemented with catalase or other neutralisers for this reason.

3. Sampling equipment compatibility

Particle counter probes, sampler heads and tubing that stay inside during VHP must tolerate repeated exposure and must not hold peroxide that later off-gasses. Electronics should be outside the isolator where possible, with only the probe inside.

Trends and excursions

Because Grade A results are almost always zero, the value of isolator monitoring is in the trend. Record results by location, shift, intervention and campaign day; review them together with glove integrity results and particle data. Any viable recovery in Grade A is investigated, with the organism identified, and the investigation considers the decontamination cycle, the transfer route, glove integrity and the interventions performed.

Related reading: hydrogen peroxide uptake in isolator filling and VHP aeration and residual H₂O₂.

References

EU GMP Annex 1 (2022), Manufacture of Sterile Medicinal Products

ISO 14644-1, Cleanrooms — Classification of air cleanliness by particle concentration

ISO 14698-1, Cleanrooms — Biocontamination control

PDA Technical Report No. 13, Fundamentals of an Environmental Monitoring Program

 Equipment
Fenggu equipment for this

FG-ISO-AF
Aseptic filling isolator

Integrated VHP bio-decontaminates exposed interior surfaces before every campaign; unidirectional airflow, H14 HEPA, pressure and particle monitoring.
View specifications →FG-ISO-ST
Sterility test isolator

Reserved VHP generator port to decontaminate the chamber and exposed surfaces before testing, reducing false positives.
View specifications →FG-PB-VHP
VHP pass box

Decontaminates the outer packaging of monitoring media and components on the way into the clean area.
View specifications →

Related application guides: Isolator and RABS decontamination · Cell and gene therapy

 FAQ
Common questions

<details>
What environmental monitoring is required inside an aseptic isolator?

The critical zone is Grade A, so EU GMP Annex 1 expects continuous non-viable particle monitoring and viable monitoring by settle plates and active air sampling throughout critical processing, plus surface samples and glove prints at the end of the operation.
</details><details>
What are the Grade A particle limits in operation?

Annex 1 (2022) lists 3,520 particles per m³ at ≥ 0.5 µm and 29 per m³ at ≥ 5 µm for Grade A in operation.
